Impact of natural killer cell’s functional reconstruction on the results of allogeneic hematopoietic stem cell transplantation

چکیده
Introduction. Currently, more and attention is being paid to possible strategies for preventing the development of graft-versus-host disease (GVHD) reducing risk infections while maintaining antitumor effect — graft-versus-leukemia (GVL). In this context, study natural killer cells (NK-cells) seems be quite promising. Aim – analyze biological functional properties NK-cells after allo-HSCT, their reconstitution transplantation factors affecting process, as well mechanisms alloreactivity NK in patients allo-HSCT. Main findings. Various types activating or inhibiting receptors, which are expressed on NK-cells, regulate functions NK-cells. Among them, main role played by immunoglobin-like receptor (KIR-receptor), mediates tolerance one’s own immune response, both directed against infectious agents. can play a decisive early relapses complications, they among first recover They also have ability eliminate recipient’s T-cells antigen presenting (APCs), thereby graft failure GVHD. There several models based KIR; however, results studies area contradictory. This review summarizes available literature data.

Functional Reconstitution of Natural Killer Cells in Allogeneic Hematopoietic Stem Cell Transplantation
Natural killer (NK) cells are the first lymphocyte population to reconstitute following allogeneic hematopoietic stem cell transplantation (HSCT) and are important in mediating immunity against both leukemia and pathogens. Although NK cell numbers generally reconstitute within a month, the acquisition of mature NK cell phenotype and full functional competency can take 6 months or more, and is i...

The introduction and evolution of hematopoietic stem cell transplantation (HSCT) could be traced back to 1950s, to the studies on interactions among irradiation, covering spleen and bone marrow from it and injection of bone marrow cells. Today, HSCT is considered a well-established, effective and promising means of therapy for various malignant and non-malignant medical conditions, both in chil...
متن کاملLineage-specific chimerism analysis in nucleated cells, T cells and natural killer cells after myeloablative allogeneic hematopoietic stem cell transplantation
BACKGROUND Chimerism analysis is an important tool for assessing the origin of hematopoietic cells after allogeneic stem cell transplantation (allo-SCT) and can be used to detect impending graft rejection and the recurrence of underlying malignant or nonmalignant diseases. METHODS This study included 24 patients who underwent myeloablative allo-SCT. DNA was extracted from nucleated cells (NCs...
متن کاملPreliminary Results of Allogenic Hematopoietic Stem Cell Transplantation of non-M3 Acute Myeloid Leukemia
Background: Allogeneic Hematopoietic Stem Cell Transplantation (allo-HSCT) is used as treatment of choice for patients with acute myeloid leukemia (AML). We aimed to evaluate the prognostic factors in 2-year overall survival of patients with non-M3 AML who underwent allogenic HSCT. Methods: This is a Cross sectional retrospective study. Demographic data and study of variables such as age...
متن کاملInvariant Natural Killer T Cells As Suppressors of Graft-versus-Host Disease in Allogeneic Hematopoietic Stem Cell Transplantation
Invariant natural killer T (iNKT) cells serve as a bridge between innate and adaptive immunity and have been shown to play an important role in immune regulation, defense against pathogens, and cancer immunity. Recent data also suggest that this compartment of the immune system plays a significant role in reducing graft-versus-host disease (GVHD) in the setting of allogeneic hematopoietic stem ...
